Genetic Underpinnings of Brain Metastasis

Summary

Patients with cancer that has spread to the brain continue to have a dismal prognosis - most patients die within 1 year of diagnosis of a metastasis to the brain. It is still unclear why particular cancer types (lung, breast, and melanoma) have a greater tendency to spread to the brain and what the genetic underpinnings are of this transformation. We will isolate DNA and RNA from metastatic tissue samples from 15 patients who have had brain surgery to remove a metastatic lesion and compare this to DNA and RNA isolated from these patients? primary tumors. To have the most yield in drawing preliminary conclusions regarding commonalities and gain-of-function in the transition from a primary cancer to one that has spread, we will take 5 patients each with the most common cancers that metastasize to the brain: lung, breast, and melanoma. Copy number analysis of the DNA and quantitative PCR analysis of the RNA will be completed by the Genomics Institute to determine which genes are upregulated and downregulated in the metastatic versus primary tumors to understand the genetic underpinnings of metastasis to the brain.
